Step 2 To a solution of (S)-3-cyano-3-({(1R,2S)-2-[(1-methyl-1H-indole-2-carbonyl)-amino]-cyclohexanecarbonyl}-amino)-propyl ester (223 mg, 0.484 mmol) in anhydrous DMF (2.0 ML) was added 1-(2-methoxyethyl)-piperizine (214 mg, 1.48 mmol), and the reaction mixture was placed in a 69 C. oil bath for 22 hours.The cooled reaction mixture was partitioned between water (75 ML) and ethyl acetate (75 ML).The organic layer was separated, washed with water (2*75 ML), dried with sodium sulfate, filtered, concentrated, and purified by column chromatography (5:95, MeOH/CH2Cl2) to give 173 mg (70%) of 1-methyl-1H-indole-2-carboxylic acid ((1S,2R)-2-{(S)-1-cyano-3-[4-(2-methoxy-ethyl)-piperazin-1-yl]-propylcarbamoyl}-cyclohexyl)-amide as an amorphous solid.
