To a mixture of 5-(((3-chloro-5-(trifluoromethyl)benzyl)oxy)carbonyl)-4,5,6,7- tetrahydropyrazolo[l,5-a]pyrazine-2-carboxylic acid (140 mg, 0.35 mmol), tert-butyl 3,5- dimethylpiperazine-l-carboxylate (97 mg, 0.45 mmol) and HATU (198 mg, 0.52 mmol) in DMF (3 mL) was added DIPEA (90 mg, 0.69 mmol). The mixture was stirred at rt for 2 h and then concentrated. The residue was purified by column chromatography on silica gel (petroleum ether/EtOAc = 1:2) to give the Boc-protected intermediate as yellow oil (120 mg, 57percent yield).
