3 -Bromo-5 -nitro-toluene, (7.7 8g, 36.0 mmol) and 1 -cyclopropylpiperazine (5 .0g, 39.6 mmol) were dissolved in 100 mL of dry t-BuOH and purged with N2 for 10 minutes. During the purge added t-BuXphos Palladacycle (620mg, 0.9Ommol) followed by sodium t-butoxide, (5.20g, 54.0 mmol) and reaction was allowed to stir at 30C under N2 for two hours. Solvent was removed under reduced pressure and the residue partitioned between EtOAc and water, then the organic phase washed with brine, dried (Na2504) and solvent removed under reduced pressure. The crude material was Isco purified on 5i02 with DCM changing to isocratic 10%/EtOAc/DCM as eluent to afford 6.75 g (64%) of JW-8a as a dark yellow, waxy solid. ?H NMR (400 MHz, Acetone-d6) oe 7.51 (t, J = 2.1 Hz, 1H), 7.43 (d, J = 0.6 Hz, 1H), 7.21 (s, 1H), 3.35 – 3.18 (m, 4H), 2.83 – 2.67 (m, 4H), 2.40 (s, 3H), 1.75 – 1.59 (m, 1H), 0.57 – 0.41 (m, 2H),0.38 (dt, J = 3.8, 2.5 Hz, 2H) ppm.
