Metal ion chelators based on 8-hydroxyquinoline (8-HQ) have been widely explored for the treatment of many diseases. When aimed at being developed into potent anticancer agent, a largely unmet issue is how to avoid nonspecific chelation of metal ions by 8-HQ in normal cells or tissues. In the current work, a two-step strategy was employed to both enhance the anticancer activity of 8-HQ and improve its cancer cell specificity. Considering the well-known anticancer activity of nitric oxide (NO), NO donor furoxan was first connected to 8-HQ to construct HQ-NO conjugates. These conjugates were screened for their cytotoxicity, metal-binding ability, and NO-releasing efficiency. Selected conjugates were further modified with a ROS-responsive moiety to afford prochelators. Among all the target compounds, prodrug HQ-NO-11 was found to potently inhibit the proliferation of many cancer cells but not normal cells. The abilities of metal chelation and NO generation by HQ-NO-11 were confirmed by various methods and were demonstrated to be essential for the anticancer activity of HQ-NO-11. In vivo studies revealed that HQ-NO-11 inhibited the growth of SW1990 xenograft to a larger extent than 8-HQ. Our results showcase a general method for designing novel 8-HQ derivatives and shed light on obtaining more controllable metal chelators.

In this study, a new bipolar membrane electrodialysis (BMED) process was proposed to replace the original resin-based method for the removal of HSS from practical lean amine liquids (from a natural gas factory of PetroChina Company). The exptl. conditions of c.d., volumetric ratio, HSS concentration, and etc., were systematically investigated. It was found that a high HSS removal efficiency of up to 95.7% with an extremely low loss rate of the organic amine of down to 1.39% can be obtained, under the optimal c.d. of 60 mA/cm2. A continuous operation verified the long-term stability of BMED for a high-efficiency HSS removal rate of up to 80.3% after a 600-h duration. From the findings in this study, we develop a conceptual exploration describing the BMED process for the separation of HSS from practical desulfurization wastewater, beyond the lean amine liquids, offering a guidance for their potential industrial applications.

Among them, chrysamide B is the most potent anti-inflammatory agent with low cytotoxicity and strong inhibition on the production of NO (IC50 = 0.010μM) and the activity of iNOS (IC50 = 0.082μM) in LPS-stimulated RAW 264.7 cells. Primary studies suggested that the mechanism of action may be that it interfered the formation of active dimeric iNOS but not affected transcription and translation. Marine natural products derived from special or extreme environment provide an important source for the development of anti-tumor drugs due to their special skeletons and functional groups. In this study, based on previous work on the total synthesis and structure revision of the novel marine natural product Chrysamide B, a group of its derivatives were designed, synthesized, and subsequently of which the anti-cancer activity, structure-activity relationships and cellular mechanism were explored for the first time. Compared with Chrysamide B, better anti-cancer performance of some derivatives against five human cancer cell lines (SGC-7901, MGC-803, HepG2, HCT-116, MCF-7) was observed, especially for compound I on MGC-803 and SGC-7901 cells with the IC 50 values of 7.88 ± 0.81 and 10.08 ± 1.08μM, resp. Subsequently, a cellular mechanism study suggested that treatment with compound I could inhibit the cellular proliferation, reduce the migration and invasion ability of cells, and induce mitochondrial-dependent apoptosis in gastric cancer MGC-803 and SGC-7901 cells. Furthermore, the mitochondrial-dependent apoptosis induced by compound I is related with the JAK2/STAT3/Bcl-2 signaling pathway. To conclude, these results offer a new structure for the discovery of anti-tumor lead compounds from marine natural products.
