The reaction mixture was stirred at rt overnight, and partitioned between methylene chloride and brine. The organic phase was washed with brine, water, and concentrated. The residue was separated by HPLC to provide compounds 20. For compounds 20f, 20g, and 20h whose syntheses involved the use of BOC-protected amine, the coupling product was then treated with TFA (0.5 ml) in CH2Cl2 (2 mL) at rt for 1 h. A mixture of 2 g (14 mmol) 2-chloro-4-cyanopyridine and 2.34 g (15 mmol) 1- cyclopentylpiperazine in 5 ml DMF was heated to 80 0C for 16 h. The mixture was evaporated under reduced pressure and the residue was treated with 100 ml IM NaHCO3 aq. and extracted with two times 100ml ethyl acetate. The combined organic phases were dried with MgSO4 and evaporated under reduced pressure to yield 3.4 g (92 %) of the title compound as grey solid, (m/e): 257.3 (MH+; 100%).

The combined organic layers were evaporated and methanol and DMF were added and the mixture was subjected to preparative HPLC purification on reversed phase eluting with a gradient of acetonitrile/water (0.1% NEt3). The combined product fractions were evaporated to dryness to yield the intermediate 3-(2-fluoro-phenylamino)-cyclohexanecarboxylic acid ethyl ester. MS(m/e): 266.2 (MH+). A mixture of 21 mg (0.08 mmol) 4-(2-fluoro-phenylamino)-cyclohexanecarboxylic acid ethyl ester, 17 mg (0.4 mmol) LiOH.H2O in a mixture of 2 mL THF/methanol/water was heated to 45 C. for 2 h and subsequently evaporated. The intermediately built acid was dissolved in 2 mL DMF and treated with 30 mg (0.096 mmol) TBTU, 24 mg (0.24) NEt3 and 13.5 mg (0.88 mmol) 1-cyclopentyl-piperazine (commercially available) and stirred for 16 h at room temperature. The mixture was directly subjected to preparative HPLC purification on reversed phase eluting with a gradient of acetonitrile/water (0.1% NEt3). The combined product fractions were evaporated to dryness to yield 6.4 mg (21%) of the title compound. MS(m/e): 374.4 (MH+)., 21043-40-3

Water (10 mL) wasadded to the mixture, which was stirred for an additional 1 h. Then,the mixture was extracted with ethyl acetate (50 mL x 3).
