After the reaction was completed, the solvent was spin-dried, extracted with EtOAc and water, and the organic phase was washed with saturated brine,Anhydrous Na2SO4 was dried, and the red solid obtained by silica gel column chromatography was the target product (13.5 g, yield: 61%).

694499-26-8, Pyridine (163 ul, 2.01 mmol) and 4-nitrophenyl chloroformate (Combi-Blocks, CatNo. OT-0341, CAS [7693-46-1]) (369 mg, 1.83 mmol) were added to the mixed solution comprising 4-((4-methylpiperazin-1-yl)methyl)-3-(trifluoromethyl)aniline (AK Scientific Co., CatNo. AK-83227, CAS [694499-26-8]) (500 mg, 1.83 mmol) and CH2Cl2 (10 ml), followed by stirring at room temperature for 3 hours. The reaction mixture was concentrated. The obtained residue was treated with diethyl ether. As a result, a target compound was obtained as a brown solid (611 mg, 76% yield). MS m/z: 439[M+H]

Step 4. 3-Iodo-4-methyl-N-(4-((4-methylpiperazin-1-yl)methyl)-3-(trifluoromethyl) phenyl)benzamide 3-Iodo-4-methylbenzoyl chloride (1.06 g, 3.8 mmol), prepared from the reaction of 3-iodo-4-methylbenzoic acid and SOCl2, was added to a solution of 4-((4-methylpiperazin-1-yl)methyl)-3-(trifluoromethyl)aniline (1.00 g, 3.6 mmol), Et3N (0.36 g 3.6 mmol), and a catalytic amount of DMAP in THF (20 mL). After stirring at rt for 2 hrs, the reaction was quenched with water. EtOAc was added and the layers separated. 694499-26-8, A solution of 2 ml trimethylaluminium (2 M in toluene, 4 mMol) is added to a stirred solution of 355 mg (1.3 mMol) 4-(4-methyl-piperazin-1-ylmethyl)-3- trifluoromethyl-phenylamine in 20 ml toluene at 10 0C under an argon atmosphere. After 1 h at r t, a solution of 436 mg (1.3 mMol) 6-(6-acetylamino- pyrimidin-4-ylmethyl)-naphthalene-1-carboxylic acid methyl ester (Step 9.2) in 5 ml THF is added and the reaction mixture is heated at 90 0C for 45 min. After cooling to 5 0C, a solution of sat. aqueous ammonium chloride (50 ml) is added dropwise. The mixture is poured into EtOAc and filtered over hyflo. The separated aq. phase is extracted with EtOAc. The combined organic phases are washed with water and brine, dried (Na2SO4) and concentrated. The crude product is purified by chromatography (SiO2, CH2CI2 / EtOH/ NH3 90:9:1) and is recrystallised from hot EtOAc and hexane to afford the title compound as a colourless solid: m.p.: 202-2040C.
